Summary

To support the growth of the Vaccine & Oncology Platforms, Celsion is seeking to hire a Scientist II, Bioprocess Development at its Huntsville, AL, facility to play an integral role in increasing its capacity and capability to produce plasmid DNA to support R&D studies.

Our lead product is GEN-1, a DNA-mediated immunotherapy for the localized treatment of ovarian cancer. Our newest program in development is a novel plasmid DNA-based, investigational vaccine for preventing or treating infections from a broad range of infectious agents including the coronavirus disease using our PLACCINE DNA vaccine technology platform.

 

Duties and responsibilities

The successful candidate will:

  • Develop plasmid DNA purification unit operations at the bench and small pilot scale (from up to 10 L bioreactor fermentation) such as centrifugation, depth filtration, homogenization, flocculation, and tangential-flow filtration (TFF).
  • Develop chromatography unit operations, including optimization, initial scale-down and scale-up.
  • Adapt and troubleshoot analytical methods such as ion exchange and reverse phase HPLC, restriction digest, and spectroscopic assays to quantify yield and purity at various stages of production.
  • Coordinate with formulation team to produce plasmid DNA of sufficient quality and quantity to support formulation development, in vitro, and in vivo experiments.
  • Draft SOPs for purification processes and analytical methods
  • Have demonstrated the ability to work independently to develop purification processes, including troubleshooting and optimization.
  • Be extremely detail-oriented and place high value in sound documentation of work for communication to supervisor, other team members.
  • Effectively communicate with technical and business-oriented audiences within the company through presentations and written reports.
  • Support other laboratory projects and initiatives as required.

 

Qualifications

To be successful in this role your background and experience should include:

  • S. with 10+ years, M.S. with 6+ years or Ph.D. with 1-5 years industry experience in purifying and analyzing plasmid DNA (preferred), RNA, or proteins at pilot scale.
  • Strong technical knowledge and direct hands-on experience with downstream processing unit operations such as chromatography, tangential-flow filtration, centrifugation, homogenization, etc.
  • Sound understanding of plasmid DNA biochemistry in microbial hosts relevant to downstream purification.
  • Understanding of engineering issues related to process scale-up (to 10 L fermentation scale or higher) and ability to apply both practical and analytical skills to implement relevant scale-down approaches.
  • Working knowledge of Microsoft Office software, particularly Word and Excel required. Experience with advanced graphing and/or statistical software such as GraphPad Prism (beneficial).
  • Excellent interpersonal, organizational, and communication skills (oral and written).
  • Some experience with technical project management relating to downstream processing.
  • Ability to prioritize responsibilities, multi-task and remain flexible with changing needs.
  • Ability to handle the pressure of meeting tight deadlines in a dynamic technical environment.
  • Experience working in cGMP or ISO quality systems and understanding of regulatory requirements for biologics (beneficial).

Duties and responsibilities

The successful candidate will:

  • Produce and characterize novel DNA formulations based on proprietary polymer and lipid based nano delivery systems using appropriate techniques including preparation of nanoparticles, agarose gel electrophoresis, DLS, and other relevant techniques.
  • Conduct experiments to evaluate cell transfection activity and stability characteristics of the formulations to advance scientific understanding of the formulated systems and achieving respective project objectives.
  • Perform quality control testing of the established formulations and coordinate their in vivo testing with Company’s project teams and external research collaborators.
  • Have demonstrated the ability to work independently to develop formulations, optimize and troubleshoot the process.
  • Be extremely detail-oriented and place high value in documentation of work for communication to supervisor, other team members and sound development of documentation practices.
  • Effectively communicate with technical and business-oriented audiences within the company through presentations and written reports.

 

Qualifications

To be successful in this role your background and experience should include:

  • S. with 10+ years, M.S. with 5+ years or Ph.D. with 1-3 years industry experience in nucleic acid formulations.
  • Hands-on experience in DNA or RNA formulation and understanding of the relevant research area are the key metrics for successful selection.
  • Experience in nano-assembly of DNA or RNA drugs into nano-complexes or nano-encapsulates and analytical techniques for particle size, zeta potential and other particle properties. Experience in cell-based assays, standard tissue culture techniques, immunofluorescence, and ELISA will be considered favorably.
  • Good presentation and communication skills and have the ability to identify and apply the appropriate methodologies and data evaluation. Candidate should be familiar with searching and interpreting relevant literature and be able to develop a focused research effort.
  • Must be able to interact with multifunctional personnel and effectively participate in a team environment.
  • Working knowledge of Microsoft Office software, particularly Word and Excel required
  • Excellent interpersonal, organizational and communication skills (oral and written).
